Book excerpt: Poly(lactic-co-glycolic acid) (PLGA) is one of the most successful polymers used for producing therapeutic devices, such as drug carriers (DC). PLGA is one of the few polymers that the Food and Drug Administration (FDA) has approved for human administration due to its biocompatibility and biodegradability. In recent years, DC produced with PLGA has gained enormous attention for its versatility in transporting different type of drugs, e.g., hydrophilic or hydrophobic small molecules, or macromolecules with a controlled drug release without modifying the physiochemical properties of the drugs. These drug delivery systems have the possibility/potential to modify their surface properties with functional groups, peptides, or other coatings to improve the interactions with biological materials. Furthermore, they present the possibility to be conjugated with specific target molecules to reach specific tissues or cells. They are also used for different therapeutic applications, such as in vaccinations, cancer treatment, neurological disorder treatment, and as anti-inflammatory agents. Book excerpt: Due to their ease of synthesis, versatile surface chemistry, ease of imaging, and biocompatibility, gold nanoparticles (AuNPs) are employed as drug delivery devices in the treatment of respiratory problems associated with spinal cord injury (SCI). We have developed a method for the synthesis of a tripartite nanoconjugate comprised of an AuNP drug carrier chemically conjugated to a transporter protein (wheat germ agglutinin coupled to horseradish peroxidase or WGA-HRP) and to potent SCI drugs. The therapeutic efficacy and biodistribution were studied in a validated animal model. Our results show that a single administration of the nanoconjugate improved diaphragmatic activity at a much lower dosage than the effective systemic drug dosage and restored the respiratory drive. The effects lasted for 4 weeks. A biodistribution study using inductively coupled plasma mass spectrometry on the same animal model shows that the nanoconjugate was successful in targeting the respiratory neurons in the medulla by detecting the presence of gold in the medulla and spinal cord. The AuNPs are also applied in triple negative breast cancer treatment. By conjugating the surface modified AuNPs with a non-soluble drug, the solubility has been increased and introduced cancer cell death after administration. The nanoconjugate structure was determined by UV-vis spectroscopy, transmission electron microscopy, dynamic light scattering, thermogravimetric analysis, and mass spectrometry.

Book excerpt: The nasal mucosa provides a rapid, noninvasive route for drug administration to the systemic circulation and even potentially to the CNS. Nanoparticles made from the biodegradable polymer, PLGA, are of great interest for use in drug delivery systems due to PLGA's relative safety and ease of surface modification. Nanoparticles may provide improved targeting and transport through the nasal mucosa. However, the optimal nanoparticle sizes and surface properties for intranasal delivery are unknown. In this study, we prepared PLGA nanoparticles within a size range of 50-70 nm containing the lipophilic fluorescent dye, Nile Red, using a surfactant-free nanoprecipitation method. The resulting nanoparticles were evaluated using dynamic light scattering and scanning electron microscopy. Nanoparticle uptake into the nasal mucosa was determined by exposing the tissues to nanoparticle dispersions for 30 or 60 minutes. The in vitro uptake of the nanoparticles by the nasal mucosal tissues revealed that the Nile Red-loaded PLGA nanoparticles were transported across the epithelial layer and accumulated in the sub-mucosal connective tissues. Nanoparticle uptake in the full thickness tissues was time dependent where 2% of the total loads of nanoparticles exposed to the tissues were measured in the mucosal tissue after 30 minutes and 4% were present in the tissues after 60 minutes. The rapid and measurable transfer of PLGA nanoparticles into the nasal mucosal tissues indicate that they may be an efficient delivery vehicle for drugs with either local or systemic activities.
