Book excerpt: Generally it is not sufficiently appreciated that electron microscopy is in fact a diffraction method. In essential aspects electron microscopes are more closely related to X-ray diffracto· meters than to light microscopes. In electron microscopes monochromatized radiation and coherent illumination (never used in light microscopy) correspond in X-ray diffractometers to the primary beam with a small divergence. Imaging ina general sense can take place in interference experiments between a primary beam and a scattered beam, or between diffe rent deflected scattered beams. This leads to the realization of an old dream in diffracto metry, namely to a general experimental solution of the "phase problem". The most im pressive analogy, however, concerns the potential of the electron microscope as a tool for structure determination (where the radiation wavelenght is smaller than the atomic distan ces). It was therefore considered timely to treat this topic in this series. It was a fortunate cioncidence that in 1976 a Workshop on "Unconventional Electron Microscope Methods for the Investigation of Molecular Structures" (sponsored by the European Molecular Biology Organisation, the Deutsche Forschungsgemeinschaft and the Max-Planck-Gesell schaft) took place, and that most speakers presenting introductory lectures agreed to publish their contributions in an expanded version in this volume. This volume is thus not a symposium report in the usual sense since it contains the majority of these introductory lectures only.

Book excerpt: This volume is dedicated to a description of the instruments, samples, protocols, and analyses that belong to cryo-EM. It emphasizes the relatedness of the ideas, instrumentation, and methods underlying all cryo-EM approaches, which allow practitioners to easily move between them. Within each section, the articles are ordered according to the most common symmetry of the sample to which their methods are applied. Includes time-tested core methods and new innovations applicable to any researcher Methods included are useful to both established researchers and newcomers to the field Relevant background and reference information given for procedures can be used as a guide

Book excerpt: If, ten years ago, one had been asked to comment on the prospects of peering into the fmest details of biomolecular organization, most electron microscopists would, I suppose at least, have been quite en thusiastic. When, during the early seventies, several groups were success ful in visualizing single heavy atoms, which undoubtedly was a techni cal triumph, this prompted the most sanguine expectations among bi ologists. In the following years, however, it began to transpire that radiation damage might impose limitations preventing us from taking full advantage of these exciting instrumental feasibilities. Fortunately, the radiation damage nightmare did no paralyze further activities, and it was in particular the work on the purple membrane which, brilliant ly exploiting the redundancy stratagem, revealed exhilarating new perspectives. Now, almost five years later, it seemed timely and appro priate to organize an international symposium to discuss and weight recent activities and current trends in "molecular microscopy". In planning this symposium, we selected topics according to our view of what is important or will deserve more attention in the near future. Taking into consideration suggestions made by the invited participants, some supplementary aspects were included; as a conse quence, the program developed somewhat beyond the scope as adum brated by the original title of this meeting (Regular 2-D Arrays of Biomacromolecules: Structure Determination and Assembly). As the meeting was organized, we had three morning sessions aimed at reflecting the "State ofthe Art".

Book Synopsis Analytical Methods in Supramolecular Chemistry by : Christoph A. Schalley

Download or read book Analytical Methods in Supramolecular Chemistry written by Christoph A. Schalley. This book was released on 2012-09-25. Available in PDF, EPUB and Kindle. Book excerpt: The second edition of "Analytical Methods in Supramolecular Chemistry" comes in two volumes and covers a broad range of modern methods and techniques now used for investigating supramolecular systems, e. g. NMR spectroscopy, mass spectrometry, extraction methods, crystallography, single molecule spectroscopy, electrochemisty, and many more. In this second edition, tutorial inserts have been introduced, making the book also suitable as supplementary reading for courses on supramolecular chemistry. All chapters have been revised and updated and four new chapters have been added. A must-have handbook for Organic and Analytical Chemists, Spectroscopists, Materials Scientists, and Ph.D. Students in Chemistry. From reviews of the first edition: "This timely book should have its place in laboratories dealing with supramolecular objects. It will be a source of reference for graduate students and more experienced researchers and could induce new ideas on the use of techniques other than those usually used in the laboratory." Journal of the American Chemical Society (2008) VOL. 130, NO. 1 doi: 10.1021/ja0769649 "The book as a whole or single chapters will stimulate the reader to widen his horizon in chemistry and will help him to have new ideas in his research." Anal Bioanal Chem (2007) 389:2039-2040 DOI: 10.1007/s00216-007-1677-1
